February 22 Stretch out your muscleDr. Hermann Gaub talked about his current work on titin kinase stretching and single DNA assembly assay at this Thursday's symposium here at UIUC. They developed an experimental scheme where the AFM and TIRF microscopy were combined such that a nano-scaled pattern could be generated by arranging single fluorescently labeled DNA line after line using a AFM tip [1]. It looked kind of cool, but on second thougnt, i feel it seems no any application or question in reality would be addressed by this apparantly teeny fancy 'nanotechnique'. Contrarily, I am very into the titin kinase work, especially when he showed a movie zooming the human muscle into the molecular level step by step.
Biological responses to mechanical stress require strain-sensing molecules, whose mechanically induced conformational changes are relayed to signaling cascades mediating changes in cell and tissue properties. In vertebrate muscle, the giant elastic protein titin is involved in strain sensing via its C-terminal kinase domain (TK) at the sarcomeric M-band and contributes to the adaptation of muscle in response to changes in mechanical strain. TK is regulated in a unique dual autoinhibition mechanism by a C-terminal regulatory tail, blocking the ATP binding site, and tyrosine autoinhibition of the catalytic base. For access to the ATP binding site and phosphorylation of the autoinhibitory tyrosine, the C-terminal autoinhibitory tail needs to be removed. Here, we use AFM based single-molecule force spectroscopy, molecular dynamics simulations, and enzymatics to study the conformational changes during strain-induced activation of human TK. We show that mechanical strain activates ATP binding before unfolding of the structural titin domains, and that TK can thus act as a biological force sensor. Furthermore, we identify the steps in which the autoinhibition of TK is mechanically relieved at low forces, leading to binding of the cosubstrate ATP and priming the enzyme for subsequent autophosphorylation and substrate turnover. Thinking a flow of water falling down freely, the atoms are clustered as the flow drops, refering to 'water drop', followed by a jet of shoots upwards. However, strikingly when a bunch of heavy small particles are dropped onto a bed of loose, fine sand, a similar phenomenon occurs [1]. Surprisingly in contrast to the liquid situation, the granular jet exists in the absence of both surface tension and cohesion. How could that be? Why the surface tension is unnecessary to play the role? Heinrich used both simulation and camera recording to investigate the system in a quantitive way. It was funny when he described how his student had dropped the camera in parallel with the granular to the top of buffer foam.."Its xx thousand dollars, so we don't want to miss that",he said so. In current issue, a PNAS paper illustrated how a high spatial resolution method named iPALM could be achieved in a three-dimenional manner in terms of improving the previous PLAM/STORM imaging.A three-layered prism was introduced to form a multiphase interferometric scheme so that the Z-position could be resolved in a much sharper way by calibrating the inteference pattern among the signals of the three CCDs. The below is the superresolution iPALM image of COS7 cell expressing the membrane protein VSVG fused to td-EosFP, rendered with z axis color-coding. The insert impressively shows two distinct and well-separated Guassian peaks with ~10nm width in Z, which obviously pushed the visible light diffraction limit by one order of magnitude. Mechanosensitive adhesions mediated by membrane proteins called integrins are believed to underlie a number of these behaviors. Forces applied either by the cells’own cytoskeleton or from external sources induce strengthening of these adhesions and trigger a variety of intracellular signals through these effects. Two papers in current issues in Science, one by Friedland et al. and the other by del Rio et al.,increase our understanding of the molecular basis for this. This two pieces of amazing work attracted me because they're addressing a fundamental question how far nonequalibrium is prevailing in such a microscopic world and how the external forces get transmitted into the cell body via the cytoskeleton so that the inside molecules can actually feel the stretch.
